A novel and effective technology for target-based screening

Target-based screening allows for rapid identification of ligands for an increasing number of macromolecular targets and is commonly used in drug discovery.

The present invention uses interaction-dependent PCR (IDPCR) to selectively identify target-ligand pairs from libraries of targets and libraries of ligands in a single solution.

By conjugating both targets and ligands to single stranded DNA, IDPCR allows for selective amplification of DNA sequence upon target-ligand interaction; the specific target-ligand pair can then be identified via DNA sequencing.

This technology is highly sensitive, easy to implement and takes place in a single solution, thus holding strong promise to facilitate and simplify both target-oriented screening in drug discovery and downstream optimization by medicinal chemistry.
