Bead-based polymerase colony microarrays and nucleic acid sequencing ("Polony-FISSEQ Beads")

Foundational work of Dr. George Church has enabled the production and use of replicable arrays of polymerase-amplified nucleic acid colonies (so-called "polonies") on semisolid support matrices (see Harvard Case #1438). The present invention extends the polony technology to provided miniaturized, high-density, bead-based nucleic acid arrays and improved methods for array-based sequencing of nucleic acids. High feature resolution enables identification and isolation of clonal sequences present on microparticles in both research and diagnostics applications.
